PERBANDINGAN MEDIA ALTERNATIF KACANG MERAH DENGAN MEDIA MANNITOL SALT AGAR TERHADAP PERTUMBUHAN BAKTERI Staphylococcus aureus

Abstract


Mannitol Salt Agar media is a bacterial growth medium that has sufficient nutrients for bacterial
breeding, but because of its relatively high price, an alternative media is needed as a substitute for nutrients of
Mannitol Salt agar. One of potential substitution media is made of red beans (Vigna angularis) which is type of
beans that is composed with high protein content. Red beans have protein content of around 22.3 grams/100
grams containing carbohydrates, fats, vitamins and minerals. Red beans are widely used as raw materials in
daily food. The purpose of this study was to determine the ptency of red beans powder to be used as an
alternative source of protein in MSA (Mannitol Salt Agar) media for the growth of Staphylococcus aureus. This
research was experimental, using the TPC (Total Plate Count) method. The ingredients used were red bean
powder with a variety of weights used ranging from 2.5 grams, 5.0 grams, 7.5 grams and 10.0 grams as a
source of protein in making MSA media. From the results of the study it was found that red bean powder could
grow Staphylococcus aureus bacteria at a concentration of 5.0 grams, 7.5 grams and 10.0 grams with the
number of colonies decreasing at higher concentrations. This was because one of the chemical compounds
contained in red beans was isoflavones which had antibacterial properties. So that at higher concentrations of
kidney beans, the growth of Staphylococcus aureus bacteria decreases.
